From the point of view of the VirtualBox manager window you want to make sure that the virtual machine has a virtual floppy drive attached in the "Storage" settings section (DOS VMs should always have one unless you changed something), and select to use your USB floppy drive with the virtual drive. Then your Mac will take care of the USB stuff and the DOS VM (and VirtualBox) will just see a floppy drive and never have to worry about the rest.
