You do not want to press the home button is if you were wanting to ask siri a 
question. 

What you want to do is take the tip of your finger and lay it on the home 
button as if you were touching the end of your nose.

touch the button then lift your finger. voiceover will prompt you to do it 
again. you will have to touch the button a few times in order for the phone to 
get an accurate picture of your finger print. 

Voiceover will tell you when a good picture has been taken.
