I had some ideas about this a while back, but didn't do anything with it. You'd need a pretty big pipe to not make the wait unbearable during backups. There are ways you could do caching, background uploading, etc. What I ended up doing is backing up to a local disk then doing nightly syncs up to S3. Works fairly well, with the downside that you can't have more stored on S3 than you have space for locally.
