I don't think it matters which meal you take it with, but I do think that the content and size of the meal can affect the intensity of nausea and other digestive side effects. I found that these side effects are minimized if I have a complete protein in the meal accompanying the Gleevec.
Weight gain on Gleevec is a common side effect. I've stopped gaining, but I don't seem to be able to take anything off. I take 200mg with breakfast and 400mg with dinner. I was on 800mg/day for a few weeks, but couldn't tolerate it.
