HI Tom, I arrived at sprycel after having organ related side effects with gleevec and tasigna. I actually felt well on tasigna, had skin rashes, hair and eyebrow loss and extreme constipation,..I could manage those side effects pretty well because I felt good. but it was the elevated liver enzymes in the 300's and overnight diabetes that moved me to sprycel. I started and have remained on sprycel 50 mgs for the last year. Had bad headches the first several weeks, but they went away. Had shortness of breath and some swelling that furosamide and potassium daily rectified. My PCR is stable at <.0002 and/or undetectable back and forth this year....I could only take half doses of any of the meds..I was o.k. on tasigna until I tried to titrate up...then my liver enzymes soared..and even after I went back down to half a dose, the liver would not calm down...Now on sprycel..I have a kind of muscle fatigue that worsens as the day progresses..I cannot climb up stairs without using my arms to support me....so the question is, will less sprycel help the cpk elevation and the fatigue? or not? what are the risks? I don't want to get resistant by taking too low a dose. Nor do I want to be doing damage to my muscles including my heart by having a continually elevated cpk... starting lower and titrating up to what one can tolerate makes great sense with these meds. taking too much and having severe side effects that bring down your dosage is very hard on the body and the organs. Initially sprycel was given at 140 but it was found to be just as effective at 100 with far fewer side effects like PE..so overall more effective at a lesser dose.
