The key command does only work if you are in the Skype window.

Will that key command work globally, or only if the Skype app has
the current focus.

how in the bloody heck did you manage to set the shortcut key?
I've looked in literally every single menu option and gone into
the preferences, and see absolutely nothing regarding this.
Quit Skype if it is running.
open System Preferences,
Press KE to go to the Keyboard&Mouse pane,
Select Keyboard Shortcuts,
Click the Plus button to the right of the Outline,
A dialogue box will appear,
Click on All Applications,
Select Skype by pressing Return,
Type Hang Up with a capital H and a capital U,
Move to the Keyboard Shortcut field and enter the key combination
you wish to use,
Note that it does not check for conflicts with other commands you
may have created,
Click Add and you will be returned to the Keyboard Shortcuts tab
and you can add more if you want to.
You may need to restart your computer for the new shortcut to take
effect.
Cheers,
Anne
