It will work without a connection but that will be pointless as you draw 
libraries and new models from the net constantly. Especially in the 
early days where new models will be scarce.
I have a machine that I do all my design work on and it is not connected 
to the internet for fear of destruction by delinquent assholes. It is 
connected to my network and then I have a laptop that is connected to 
the internet. On the laptop I get all the latest updates and then I run 
Syncback to update any libraries on the development machine.
