previous discussion with a new subject.

This one is more complicated than would initially appear. Ideally you would
want to setup the auto-reply rule on your mail server so that your computer
does not need to be left on and processing mail. This isn't always
available/possible, though. Also, in constructing your rule, you want to try
to avoid mail loops (where your auto-reply triggers someone else's
auto-reply, which then triggers your auto-reply again and so on) and
auto-replying to mailing lists (A big no-no). Lastly, you probably don't
want to auto-reply to the same person more than once telling them you're
going to be out of the office for 2 weeks.

First off all, create a group in the Address Book called "Replied Senders".
Then go to Tools -> Rules and create a new rule (POP or IMAP, depending on
the account type you receive your mail on) with the following:

If
Execute if all criteria are met
Sender   Is not in group   Replied Senders
Specific header    Return-path    Is not
Specific header    Return-path    Does not contain    MAILER-DAEMON
Specific header    Precedence    Does not contain    Bulk
Specific header    Precedence    Does not contain    Junk
Specific header    Precedence    Does not contain    List
Specific header    X-Autogenerated    Exists

Then
Reply    Reply Text...
Add sender to group    Replied Senders

Just make sure and delete the addresses in the "Replied Senders" group
whenever you turn the auto-reply rule off so that, the next time you turn it
on, you will be starting fresh.
