E-TTL fires a preflash to perform some magical

The litelink will fire on the preflash instead of the real one.

Does this mean you intend to have 2 420EX's? One for the shoe and
one for the litelink? It is not a problem if the popup flash is the
master and the 420 is on the litelink.
I'm not sure what body you have, but on mine I know that manual
mode uses regular TTL, and other modes may as well. Program
mode is ETTL, I'm not sure about Av and Tv.
I understand the 550EX has a switch to turn it off, but the 380EX
does not and I don't think the 420EX does either.

If the slave is much stronger than the popup, yes. Either much more powerful
or much closer will do. You'll probably have to experiment to find out what
the cutoff is for your combination of flashes. Of course, if the slave is that
much stronger than the master, you just have a wireless triggered single flash.
I always use the 85/1.8 for portraits with the litelink, so I haven't had
to worry about it all.
