I don't think pdf's know anything about double sided printing.  However, it
is much more common for the music to begin on page 3 than page 2.  You might
consider putting the instrumentation on page 2.

I strongly suggest going with wire coil binding instead.  Combs are more
likely to bind, tear the paper, get crushed, and make noise while turning
pages.  Also, wire coils allow the score to be opened a full 360 degrees,
which can be handy.  No contest in my opinion.

Not sure because I wouldn't trust the job to them.  I have found it's enough
of a challenge just to get a print shop to trim and bind scores properly.
