Um, no. PlusOptimizer is an HFS+ formatter that doesn't actually loose
your
data.

Here, an excerpt from the read me of PlusOptimizer:

Optimizing Your Startup Disk

Your startup disk is the disk containing the current System Folder.
Since the open system files on the disk containing the current System
Folder cannot be defragmented, you need to start up from another disk to
defragment these files and completely optimize the disk. If you do not
have another hard disk with a System Folder, you can startup from your
Mac OS system CD. To optimize Mac OS Extended (HFS Plus) disks, the Mac
OS system CD must be Mac OS 8.1 or higher.

Directory Damage

PlusOptimizer performs an examination of the disk directory data
structures on which it relies. If PlusOptimizer finds that your hard
disk directory is damaged, try running Disk First Aid (supplied with
your computer) to see if it can repair the damage. Because Disk First
Aid does not check for every type of directory damage that PlusOptimizer
checks for, it is possible that Disk First Aid will not be able to
confirm the damage. PlusOptimizer may be unable to optimize your disk
until the damage is repaired. If necessary, use Alsoft's DiskWarrior to
repair the directory damage.
