Here is the basic differential diagnosis ladder for OS X, it's really simple.

1) Start in Safe mode. In all versions of OSX this starts you with a minimal 
set of Apple drivers, eliminating issues with third party drivers, and in 10.4 
and on, also clears out many caches that OS X maintains, like cached library 
links, fonts, etc. It's entirely possible that this will fix the problem.

If this does not fix the problem:

2) Create another admin user. (or have one created from the beginning for 
diagnosis use only), and log in as that user.

2a) if the original issue exists, then it is a systemic problem, fixable by 
solutions like archive&install or re-applying the latest combo update.

2b) If the original issue isn't repeatable under the new user, then the problem 
lies in the user directory somewhere, and things like A&R or reapplying updates 
will do no good, the issue lies in the user preferences, add-ons or application 
support files.

3) If all of these do not fix the problem, then the issue is likely a hardware 
issue: bad RAM, a failing drive, etc.
