With this setting, no other security measures will have any effect whatsoever and you are a wide open relay.  The ONLY non-open relay setting possible with IMail is to use SMTP Auth (which you have attempted but thwarted) and to use Relay for Addresses.  If your users can't send under this configuration, then either the addresses are being entered into IMail incorrectly, the user's client software is configured incorrectly or the network is configured incorrectly.

To relay for your 192.168.100.xxx block only, you need to select relay mail for addresses and in the addresses dialog add a group of computers.  In the IP address enter 192.168.100.0 with a subnet mask of 255.255.255.0, repeat for each block of addresses on your network, example:

192.168.100.0 255.255.255.0
192.168.101.0 255.25.255.0
192.168.102.0 255.255.255.0

Start and stop smtp services and then start working out why your users can't send mail if they're still having trouble.
