I have seen a few issues with the Imail 8.01 and Declude regarding conflicts SPAM filtering. Has it been determined what is the best setup?
There aren't any known conflicts, actually -- you can use either one or both together (with some obvious exceptions; for example, if you set IMail v8's anti-spam to delete an E-mail, a whitelist in Declude JunkMail can't override that).
I believe the issue being reported was back with the original IMail v8 that would block E-mail by default, even when it appeared to be turned off. So people would upgrade to IMail v8, turn off the IMail v8 anti-spam, and find that Declude JunkMail was blocking messages that couldn't be found. It turned out that the IMail v8 anti-spam wasn't completely disabled. Now that it does not block by default, this is no longer much of a concern.
