Thank you for your advice.

I could hear sound changes caused by the Calf Filterclavier by adding a 
midi track, inserting some notes in this midi track, connecting it to a 
Fluidsynth instance, then selecting the Out 1 track in the MusE file, 
then there adding the Calf Filterclavier synth by right-clicking one of 
these grey "empty" buttons.

I set up a song the same way as described above, this time with the Calf 
Rotaryspeakter synth, and I could hear sound changes by this synth.

I have a proposal:

All LV2 synths which do not work (have no sound changing effect) by 
adding them to a song by right-clicking that grey area in the left pane 
of a MusE song ("Add synth" => "LV2 synths") should be removed from that 
list of LV2 synths there. Otherwise users will wonder why this and that 
LV2 synth does not work. And only those LV2 synths that work if added 
via the grey "empty" buttons in the Out 1 track should be listed in that 
other list.
