It can be difficult to move one of the default directories to another location, so I would be hesitant to do try to move ~/Movies or ~/Music. However, it is very easy to put the actual movies in another location, and to point iTunes to another location, which is what I do. I would not hesitate to put other user's home folders on a different drive, but I would only do that on an internal drive.It is trivial to do, you just right-click on the user in the System Prefs and choose advanced options, then change the path to the home folder.
