It works for me in a G:\MOZPROFILES dir. Since there seems to be problems with your setup, might I suggest that you delete Mozilla completely, reboot. Set the MOZILLA_HOME variable to a new existing directory, reboot and then reinstall Mozilla. See whether it catches the new profile directory when it sets up a new or migrated profile for you.
