If you're looking to extend the wiki to an extranet type setup there's a couple of different projects worth looking into. First up is a pretty lightweight (resource wise) setup in Dokuwiki, which comes with ACL's as standard. Then there is the much chunkier but certainly more featureful Plone. I'm a big fan of Drupal but as a previous poster mentioned, each tool has its place, Drupal is a wonderful CMS, not so good as an extranet setup.
