you only need to backup your tripwire signature files. this database
files that contained signatures or checksums of your system. once
backuped, remove them from that box and keep them offline. whenever you
want to check your system just insert the diskette that contained this
checksums.
another way to do it keep the checksum files inside your lan, assuming
the server where you have installed tripwire is on dmz, and let your
tripwired server connect with this inside server using ssh.
sorry can't help much on the details. it's been a long time since i've
down this but it is one of the higly recommmended ways.
