With all due respect I think Ian's advice gives the repro people a 100% certainty of not producing the best result. Sharpening should only be done once and should be done last. It should be done when the repro size and viewing distance is known. I doubt very much whether a 300Mb file is needed. As this question has cropped up before would it be worthwhile contacting a poster printer and asking them to describe their technical specification to post onto the Annexe?
Although not related directly to digital capture the last 48 sheet stock image I sold had to be re-scanned from a 35mm tranny. I already had a perfect drum scan of sufficient size. The reason the new drum scan was necessary was because of the way that this printer was doing the printing of the separate sheets. Specialised equipment requires specialised files.
Many small digital files have been used as large posters. I think the best course of action is to ask the agency for the spec. required.
FWIW BBC Online used the image of John Prescott punching a heckler on a 48 sheet poster. It was a 4Mb file.

I havent got the 1ds but make sure you get the interpolation actions
from fred mirandas web site and interpolate with sharpening. It will
probably give you a 300mb 8 bit file, that should be fine then. At least
then you wont have the repro house resizing the smaller file and doing
something wrong.
