Anything you do to a clip, with the possible exception of clip gain, is a destructive, file based process. Panning of the type you described would definitely be an automation process. You'd have to set that track to Auto Write for the initial pan position, set it to the new position where you want it to change and engage the transport for at least a moment to have the change recorded. Under certain circumstances, you might have to choose "Write to Selection or write to End, etc. It depends on the specifics of the panning, location, whether there's other panning on the track later on that needs to be preserved, etc.
