Ah, gotcha. The answer can vary, depending what you use to create your icons. Here's my particular setup. I use ImageReady to create 24 bit PNGs with transparency to use on Mac OS X and 8 bit PNGs with transparency to use on Windows. Then I use a little program I wrote to extract each PNG's mask to a separate file (alternatively, you can use the PNGUtilities plugin to load the PNGs with their masks).
