If anyone is looking for tissue for a bank, I have a boxes and boxes of derm 
specimens and GI specimens. I know they're probably not what everyone needs but 
if someone out there does need them, you can have them. figured I'd ask before 
I disposed of them... at the landfill next to the city water well... LOL, just 
kidding, I know someone out there will not think that's funny, it's just a joke.

Hello all,



returned late last night from Pittsburg and the Intel-International Science and 
Engineering Fair, May 13-18.  The largest most prestigious pre-college STEM 
fair in the world. Took  four high school students from the Spokane region.  
One of our other "Team Washington" students from near Seattle won the second 
place prize for the entire 1,400 project fair from 81 countries and regions.  A 
$50,000 scholarship with a chemical engineering project.


While there, I viewed or walked by every project.  Saw 50-60 with histology 
images/histology projects.  IF, IHC, H, Safrinin O, FISH, PAS, decals, 
frozen, TMA's; anything and everything we all know.  On osteoarthritis, breast 
cancer, kidney disease, diabetic pancreas, neuro, lymphoma's and more.


If you are able to help what could be a potential world class student in the 
field of histology/pathology/medical and clinical research, hope you will.


Sorry I was never able to get to the local histology school while my students 
were being judged in Convention Center.  Couldn't seem to ever contact them.

Hey Alan,


I use a Airfiltronix Ductless Bench Top Hood and it meets all my needs.

I gross about 7500-8000 GI biopsies per year and do any hand cover slipping 
under this hood (which is rare if I can help it).


The unit is made of clear acrylic, is see thru and is framed out in light 
weight aluminum. It has a clear Velcroed thick mil roll up sash which is very 
easy to work with. The system has two blower units on top. Both units have 2 
inch carbon filters that I purchase from Mercedes Medical and replace twice a 
year.


I did add a grated plastic secondary containment box which I bought through 
Grainger  to place the unit on so that I could keep my formalin cube under the 
hood and not worry about spillage. This also elevated the work surface off the 
counter by 8-10 inches so you are not leaning over the work and therefore 
saving your back.


I had this unit tested by an outside agency along with the whole lab before we 
started operation and then again after we started operation. Our PEL numbers 
for Formalin, Alcohol and Xylene were barley negligible. I have used the badge 
system every year since the original testing  with the same results. This to me 
is the key to selecting any hood, does it keep the PEL numbers within exposure 
limits.


The purchase price is around $2800.00 + filters @ $180 ea.

I am in a 8 Physician GI Laboratory and our Medical Director is from Inform DX 
(IDX) (formally Miraca Life Science).


The consulting staff at IDX states with the Medicare reimbursement drops over 
the past couple years, that unless you run a high volume (>15-20/day) of 
H-PLYLORI IHC, it is not cost effective to do the Vantana or Dako  IHC stains 
as the reagents are just so expensive and do not have a very long shelf life.


We use a special "HP-Blue" stain from Anatech LTD out of Battle Creek, MI.  The 
stain is cost effective as it can get pretty good results for  the cost and we 
are able to put it on our automatic stainer and run a program just for HP Blue. 
We then make an addition charge for the special stain slide.


If a diagnosis can not still be reached from the HP Blue special stain and an 
IHC is still required by the pathologist, we send it to IDX's  Larger Lab where 
they can complete the IHC stain and they will charge the patient accordingly.


StatLab has some hand staining IHC Kits and that is more cost effective while 
still achieving the same results as Vantana or Dako. We are just now looking 
into this option for cost effectiveness vs. results quality.

I also have used Sakura Smart Write (Primera printers repackaged under the 
Sakura name)  slide and cassette printers since they came out. I had some 
issues with the slide printers in the beginning but they have worked out all 
the bugs and greatly improved the machine. I do very little maintenance other 
than clean rollers on the slide printer and never had any issues with the 
cassette printer. I recommend them for Small Labs for sure. They have multi 
cassette holders for larger labs.

Hi all,


Does anyone have any experience with embedding an acrylic prosthetic lens for 
EM, or any other way ? We have a case where they wanted EM on a lens to 
identify a  specific bacteria. We have never done it and it would take some 
work to figure it out. Also, I told them that unless there is some morphologic 
marker for the bacteria, EM would not help much identifying it.

Any ideas?

Any labs that handle this kind of thing? They are willing to send it out.

I've used the Primera slide printer since it came out.  I like it a lot.  It 
needs it's maintenance kept up like anything else but other than cleaning the 
rollers there is very little else that needs cleaning.

I totally agree with your assessment of the Slidemate printers. Have not had 
any experience with the newer model.
Applying slide labels shouldn?t be an issue when dealing with one block and 
slide(s) at a time.  Some of this could depend upon your software available to 
you. Our system generated just the labels needed for the block you were 
cutting, on demand. You should be able to get a nice Zebra printer for under 
$700.

Another of our labs used the new Sakura/Premera printer and really like it.

We have used the older version Slidemate slide labelers from Thermofisher for 
the last three years.   I think putting the patient information directly on the 
slide is preferred however the wear and tear on slide printers means higher 
maintenance costs and more down time.  Thermofisher has a newer version of the 
Slidemate now that has been designed so that there is less wear and tear on the 
machines.   Currently when one of the old labelers breaks down we have to send

My lab was set up new in 2012 with negative pressure air flow with 100% make up 
air through the HVAC system. The contractors used current Laboratory 
ventilation protocols as I understand it and it included negative pressure 
throughout the lab.


The purpose is to prevent the fumes from Formalin and other chemical in the lab 
 from seeping out from under the doors and through wall cracks or windows to 
other populated areas exposing others to known carcinogens and fowl smelling 
fumes such as xylene and alcohol.


I still have to work under a hood to protect my self from formalin exposure per 
OSHA protocols.

I am exploring digital pathology and I am unclear about the current CAP 
requirements and how those may change if the platform is FDA approved for 
primary diagnosis.  Is the wash out period still necessary if it?s not 
considered and LDT?  Does the personnel still have to be qualified as high 
complexity testing personnel to scan the slides?  Any help or other information 
is greatly appreciated!

Hey folks,



I am looking to purchase some blocks with positive H pylori tissue.

I have never done this before as we always had plenty of positive tissue in the 
wet tissue room. Has anyone had to buy tissue?

If you have, who did you find to be good to work with and how is

it sold, by the block by the case by the pound ( LOL). Really could use some 
direction on this. I just cant keep enough acceptable positive H pylori control 
slides on hand and am always running out. Would like to have a 6 month supply 
to work from and be able to keep a head with.

Hello,
Can anyone recommend a band saw for pathology? We are looking 
for a new band saw for use in necropsy and research. It has to be a "meat" band 
saw, as workshop band saws aren't compatible with wet tissue.
A table-top model is preferred, and cheaper than a floor model. (Plus, the 
floor models all seem to come with a sausage-maker, which is pretty gross.)
If you have used a table-top band saw in your pathology lab, 
I'd appreciate hearing from you.

In our GI Laboratory we have both and we do between 35 and 55 blocks per day. I 
would not want to have to choose between the both.



I would get a "new" Auto stainer because the quality of your stained slides 
would be more consistent and be easier to assess for QC purposes.



I would get a "used" Tissue-Tek coverslip machine because it is a work horse 
tank and rarely has problems and would increase productivity by 5x over doing 
hand coverslips as well as reduce the chemical hazards exposure to the Tech.

I have seen so many more problems with auto coverslippers, they break down, the 
cover does not hold up over time for archiving, etc., that I would would 
probably chose an autostainer.

I started a lab with the help of Miraca Life Science. Policies and procedures 
is probably the toughest. Finding a vender to take xylene waste an getting haz 
waste credentials. CLIA certifies  & monitors the lab. I used SharpsInc.com for 
my biohazard as my volume was to low to use stericycle and not cost effective. 
doing everything yourself is not easy but can be done but your output is low 
due to the amount of taskes to be completed.

Ann,

I would recommend validating manual microtomes more often then automatic. Any 
traditional gear driven microtome does change overtime as gears wear. This 
eventually will lead to the micron indicator on the microtome not reading what 
you are truly cutting. Automated, microtomes driven by stepper motors tend to 
remain accurate longer but you may want to document the accuracy every couple 
of years.

To check calibration most service engineers will mount a micrometer to the 
microtome and measure the feed of the microtome as the hand wheel turns. The 
procedure generally takes 5 minutes. Check with your microtome manufacture to 
see if they can assist you.

I purchase 5 liter cubes from Mercedes Medical.
They can provide test results on each batch of water and fax to you.

I did have a CLIA inspector advise to document daily on the water as is
written on the cube for clarity of water, no sediments or signs of 
contamination.

Think 5 Liters is about $12.50 and I use a little less than two a month.

Hey Histonetters,



I don't endorse products but this one is really good.



I don't get a kick back and no relatives work for the company, I just wanted

to give you all a good product to use if you are looking. I looked up the 
company

and found they are the last U.S. Made Rubber Band company and based out of

Arkansas.



I think I have had 2 eye injuries and multiple painful mishaps from

rubber bands that break far to easily so from that perspective I feel

I can give this product a thumbs up.



For the last 4 years I have been using  Alliance Rubber Company

non- latex rubber bands. The size 64 (3.5 x 1/4) work best on Slide folders

I have found. The Orange color rubber bands are easy to see and

hold the file folders together great and they rarely break.



I know it sound silly to talk about rubber bands this way but I was

really getting tired of being snapped by faulty rubber bands when trying to

wrap slide folders or paperwork.



I just had my first rubber band by Alliance snap a couple months ago which 
caught me

off guard cause it had been so long since that happened. It was then I look to

see what brand we were using and it turned out that we had been using the same

NON-latex rubber bands from the Alliance Rubber Company since I opened the 
Laboratory in 2012.

Before that at other labs breaking rubber bands were just part of the daily Job.



I think purchasing has gotten their best price on Amazon but they also use 
staples and

Office depot for supplies. I informed them that I only want these rubber bands 
and they

can get their best pricing where ever.



My fingers hurt a lot less after distributing slide folders and I don't have to 
swear as much. Ha Ha.

Funny how little things can really bother us.

I am in agreement with Carlos except low operating temperature
can effect the lubrication of the slide mechanism by gelling up and
causing the slide mechanism to skip and jump.
Lubrication of the internal mechanics of the Microtome are very
necessary on a regular basis. If volumes use is high then lubricants
dry out causing skipping and jumping of the slide.

Either way my recommendation is to have it Serviced right away
and do not use any more until you have done so.

[Histonet] Price Gun

I ordered mine on ULINE.com and got it for $128.00 they also supply
the sticker rolls. This helped me as I get standing orders once a month.
I apply the received sticker next to the containers exp: date. When I open
the container and only use a portion. I circle the EXP: Date, and Received date 
sticker
in Red and hand write the date it was opened.

This has expedited being able to put supplies away and looks a lot cleaner
during inspections.

Hi Marcus,
As stated previously by others there should be no problem putting the Eosin 
into your sections but I would suggest that you do not leave out the endogenous 
quenching so you can show your red blood cells. The reason being that there are 
many other cells that may then show up as positive. Instead I suggest using an 
Eosin/Phloxine mixture which nicely highlights the red cells differently to the 
rest of the general matrix material.

Does anyone have a processing cycle that is good for fatty tissues like breast 
that they would be willing to share?  This will be used on a VIP5.

Histology Technician

The purpose of the Histology Technician is to provide the Pathologist with
diagnostic slides to enable them to make a diagnosis.



Duties:

?  The main duties are: embedding with correct orientation, cutting and
staining of histology specimens

?  Correctly prepares special stains and solutions

?  Performs immunohistochemical stains

?  Daily maintenance of laboratory equipment

?  Early morning shift, Monday ? Friday at this time, possible Saturday
shift assignment or rotation



Qualifications:

?  1 year previous experience as a Histology Technician

?  High school diploma or equivalent

?  ASCP certification is a plus

Need opinions on this matter.
What is the back up plan in your institution, for slide printers and
cassette printers?   Are you going back to handwriting slides and putting
a label or using  xylene resistant labels directly on to your slides.

Karen,

We were hit with minor incident by CLIA for not having Temp and Humidity 
documentation as well.

We also use the equipment manual to set the Temp and Humidity operation ranges.
WE have a hi/ lo Temp and Humidity monitor on the door jam between both rooms.
We hand document the time and the Tech signs off we * any day we have out of 
range
and write down the corrective action which is usually notified maintenance.

We also only document during hours of operation.  Histology does not run 
24/7/365 like the clinical Lab
so we have to adapt some of the rules to our operation.

Our block file room has a back up A/C unit so

I know that there are many people out there that have a passion to teach what 
they know and we would very much like to put a call out to those who would be 
interested in presenting a workshop or two at our 2016 Florida Society for 
Histotechnology Annual meeting in the beautiful gulf coast area of St. 
Petersburg FL on May19-22, 2016 at the St. Pete Bayfront Hilton. We will have 
two and half days' worth of lecture time, starting with our popular 
Administrator and Supervisor Forum, the whole day HT preparedness workshop for 
students or those who are planning on taking their HT/HTL certification through 
ASCP. Please consider this great venue and submit your abstracts.

We are interested in topics on special stains, IHC, In Situ, Lean, billing, 
safety, digital pathology, ergonomics, cytology, CAP/CLIA inspection tips, 
leadership and any other topic that is relevant to today's histology 
laboratory. Please send abstract to John Shelley at

Good Morning,
I have been asked to monitor the room temperatures in the Histology lab, and 
anywhere we store paraffin blocks, slides and reagents.  Some of you I know 
already do this.

1st Question:   What is your temperature range?
2nd Question:  Are you physically monitoring it daily and writing it down on a 
temperature chart?

The reason why Question 2 was I purchased these new thermometers that I can put 
in the Hi and Lo.  It also has where I can download it on to a scan disk so I 
can down load and put on a flash drive or store in the computer.  I am not sure 
why these will
