Dear members of the ccp4bb,
I have a somewhat unusual question. For many years I have shown the movie 
“Naturally Obsessed - the making of a scientist” for educative purposes in my 
classes at the faculty of medicine in KU Leuven. I just noticed that the movie 
is no longer freely downloadable. 
https://pbsinternational.org/programs/naturally-obsessed/ PBS responded to me 
that they no longer have the distribution rights. Could anyone point me toward 
a downloadable movie or perhaps a DVD so I can show it in my class this 
semester?

A PhD position is available in the Laboratory of Prof. Chris Ulens, KU Leuven, 
Belgium (http://www.xtal.be<http://www.xtal.be/>). The research in the 
Laboratory of Structural Neurobiology is aimed at understanding the molecular 
mechanism of ligand-gated ion channels. Our methods rely on X-ray 
crystallography and cryo-EM in combination with electrophysiological recordings 
and fluorescence-based techniques. We have access to an integrated 
macromolecular crystallization facility with a Mosquito crystallization robot 
and two RockImagers. We have access to a 200 kV Glacios microscope on campus as 
well as 300 kV JEOL CryoARM300 at the nearby EM facility in Brussels. Current 
projects are situated at the crossroads of structural elucidation and drug 
discovery of different pentameric ligand-gated ion channels.

We would like to recruit a PhD student with prior experience in purification, 
cryo-EM or X-ray crystallography of membrane proteins. The position is open 
immediately and applications will be accepted until the position is filled.

the molecular mechanism of ligand-gated ion channels. Our method relies on 
X-ray crystallography and cryo-EM in combination with electrophysiological

macromolecular crystallization facility with a Mosquito crystallization robot 
and two RockImagers. Current structural targets include the family of 
pentameric ligand-gated ion channels (Cys-loop receptors), TRP channels and CNG

expression, purification and crystallization of membrane proteins. Prior 
experience with X-ray crystallography or cryo-EM is considered a major

I would like to get recommendations for a proper cryo solution for a  
crystallization hit from the Hampton crystal screen Ammonium di- 
hydrogen phosphate 0.4M. I tried increasing glycerol up to 30% with  
the same ammonium phosphate concentration or increasing glycerol up to  
30% in the presence of 1.3M ammonium phosphate. Both gave iced up  
drops (I only tried quick and dirty tests by dipping a cover glass in  
liquid nitrogen).

crystallographers see potential benefits from the functionality of the  
iPad to swipe and pinch through drops. We are looking for specific  
comments from Formulatrix users, but other users are also welcome to  
comment.

I understand you want to use in situ diffraction for assigning scores,  
but I guess it is still useful to visually inspect drop images before  
you decide to go to a synchrotron, no?
So let me rephrase the question and ask whether users see any added  
benefit of using an iPad for decision making on 'going to a  
synchrotron/buying Marcus's PX scanner' or not.


Thank you for the private posts indicating we now have a good excuse  
to buy iPads from research grants. Unfortunately for the PI's, we may  
have to buy them for all lab members now.
Would also be interested to find out if there are any members of the  
crystallography community that have experience in writing iPad apps.

Our research focus is on ion channel homologs from different classes.  
Our strategy to identify targets for structural studies relies on a  
GFP-based pipeline for pre-crystallization screening and nanobody  
technology for purification of GFP fusion proteins. This pipeline has  
been established for E. coli and insect cells and permit scale-up to  
50 liter cultures in Wave bioreactors. Our laboratory hosts an  
integrated macromolecular crystallization facility with a Mosquito  
crystallization robot, 2 RockImagers and an in-house rotating anode X- 
ray source.  In combination with electrophysiological recordings of  
recombinantly expressed ion channels in Xenopus oocytes we aim to  
understand the relationship between structure, function and  
pharmacology of ion channels.


We have ongoing collaborations with Pharma Inc. to aid the rational  
design of new drugs and to facilitate treatment of disorders related  
to dysfunctional ion channels. Our laboratory is also a participant in  
the FP7 consortium Neurocypres devoted to structure, function and  
disease of cys-loop receptors. http://www.neurocypres.eu. We have  
lively interactions with outstanding groups on campus, including the  
ion channel group of Thomas Voets (TRP channels) and the structural  
biology group of Sergei Strelkov.


Prior experience in cloning, cell culture and protein purification

Compensating like this is of course not the best (go and recollect!)
but still way better than unusable data in the meantime. In the case
Sergei originally described it would at least indicate what the
problem may be. Sergei did not say which detector was used for the
data collection so we don't know if it was a Pilatus or a CCD. Maybe
Sergei can fill us in on the details?



The data that Sergei described were collected by yours truly at
beamline ID14-2 of the ESRF, so detector is a Quantum CCD. I collected
data for another protein crystal a few hours before with 0.5 degree
oscillations - no abnormal statistics here, so I don't suspect cryo or
loop instabilities are the underlying cause as suggested before.
The choice of 0.1 degree oscillations appears somewhat unfortunate in
retrospect, but this is what several programs (Best/mosflm/DNA)
suggested to reduce overlaps. The data were collected for the only
crystal that showed decent diffraction to ~3A out of more than 30-40
screened.

I'm trying to install a nVidia Quadro FX3000 card on a PC running  
Ubuntu 8.10 intrepid.


I run into the following error when I install drivers I downloaded  
from the nVidia site:
The installer fails to find a kernel interface from the nvidia ftp  
site and fails to compile the kernel.


Any help would be greatly appreciated.
