Growing Exchange IS Out of control

I have an Exchange server growing uncontrollably.  A full backup ran last night and purged the log files so far today the transaction logs have grown 9 GB.  I have looked through the users expecting to find a run away mailbox but nothing out of the ordinary there.  Any ideas where I might want

trouble sending to specific domain

From time to time my users receive the error listed below when attempting to send mail to that specific domain. Occasionally the messages get through without issue and other times not at all. We do not have problems sending mail to other domains. Any ideas? Help would be greatly appreciated.
