`*While* you're using it? No, never, because there will
always be a discrepancy between your last backup and the
current contents of youyr folders, unless you backup every
time that you have received or sent mail.

(I have written a small batch file that backs up my daily
changes, encrypts them, and stores them on a different
partition. You can't expect more from a backup. Yet, one
needs to make regular backups on a different medium too - a
tape, a cd, a removable hard disk.)
