I do not see how this could be one in OWL, but you can do it in SHACL.
ex:SingletonClas a owl:Class;
a sh:NodeShape;
sh:path [
sh:inversePath rdf:type ;
] ;
sh:maxCount 1.
This would not prevent it having additional instances from subclasses as the
constraint only deals with the direct instances. You could do something more
complex with paths to address possible indirect instances if this is even a
consideration.
