If I understand what you mean correctly. Do you mean like use the Flash drive  
in place of and SSD or HDD, and do a proper install onto it?

Would I need two flash drives one with the installation iso, and another  
which I would install onto?

I don't know how to do this, if this is indeed what you mean.
