Thanks everyone. So, if I deactivate "maintain sync" then it seems to stop the glitches. After about 15 minutes it started to sound slightly out of sync. Both RPi are now connected to Powerline and I'm using a third Pi as server to make sure it's not an issue with one instance of Squeezelite running locally and the other over network.
