It may be possible to get better compression on a file that contains
lots of pixels of the same colour, but all pixels require the same basic
storage, regardless of whether they are transparent or not!

Sorry I should have been specific - my above comment is based on my experience of exporting 8-bit PNGs from FireWorks. When I made a large area transparent (the background area around the text for a navigation sprite) it gave me quite a significant sizer difference compared to a filled background.
