I have my mibs in several different directories and have never had an issue.  
But I always do my zenmib run from the command line as the zenoss user.

To address question number 1, what you have to do if you want to proactively 
monitor the interface status without relying on traps, you will have to create 
a datasource, and then create a threshold.  Zenoss will still show the 
interface up on the OS tab, but you at least get your info without relying on 
traps or syslog.  Take a look at this thread for more info:
