I would take the filter off for the purpose of testing it out, just in case
it is on the wrong one. Just put it back when you are finished sorting out
the problem.

Assuming it is a lack of water, my suspicion would be that the pipe
kinked/blocked just where it goes through the gap into the boiler section of
the machine. Switch the machine off and uplug from the wall. Remove the
covers and follow the pipe from the tank, check where the pipe goes between
the gap. Take the pipe out off the gap, switch the machine back on and try
pulling a shot to see if it can build pressure with the blank portafilter
installed. If it does then you know where the problem is, otherwise unplug
again and follow the pipe checking for any other kinks in the pipe.

My Brewtus IIIR had an issue with the supply pipe kinking, when pulled
slightly, and slowing water to the rotary pump when I was pulling shots.
