I installed Avast! antivirus http://www.avast.com/ on my wife's machine when I recently reinstalled Windows XP Pro on her system, and it has been great. As far as Anti-Spyware software goes, I use the multi-layer approach and install Spybot S&D, AdAware, and MS Anti-Spyware. I run weekly scans with Spybot and AdAware, and MS Antispyware runs all the time.
Of course, I also keep all machines up to date with patches, etc., and I have fairly aggressive anti-Spam settings on my mail server. A reasonable amount of paranoia has kept our household computer virus-free for...well...forever (knocks wood).
