Assuming that you mean when a Windows clients prints using a UNIX system
as a print server, that depends on how you specify/find your printer.  If 
you
browse for a shared printer you may find it via Samba but SMB isn't a native
CUPS protocol.  I believe that CUPS natively supports IPP, JetDirect and
LPD.  You can use IPP directly from Windows by specifying a local printer
on a  TCP/IP port.
