It's not a command. It's a line to be put into e.g. a
/etc/modprobe.d/speakpu.conf file. You can check whether it got
interpreted correctly with

Speakup does not do such thing out of the blue.
I have no idea what is happening with your device, but AFAIK only you
have reported this, so I have no idea what's happening.

You can try and recompile with putting 86 everywhere there, you will
most probably not get any difference, since speakup has no reason to
change the volume out of the blue.
