That is the only point of contact and that is where the grease goes. I assume the grease just helps heat transfer between two contacting pieces. As for a suitable amount. The heat sink and cpu are squeezed together by the heat sink clip so too much grease is going to get pushed out the sides. I would say just enough to coat either surface.
