I'm also experiencing the same problem. After upgrading to Lion, it appears that Growl is intermittent. It will work for a while then stop working. Also, I'll get sounds from notifications at what seems like random times, or maybe when the notification is supposed to be appearing, but I cannot see it.
Also, the Preview does not work unless I completely kill Growl and restart it (i.e. clicking Stop Growl does not actually stop the daemon, I have to manually "kill" it from a Terminal window). After a fresh start, the Preview works until I leave the System Preferences. In addition, notifications start working for some time, then randomly stop.
