Do any of these editors support an "ISPF-like" hierarchy of directories? For
example, on open look in \aaa\x first, and then in \aaa\y, and finally in
\aaa\z - but always save the changed source code in \aaa\x?

Is highlighting available for Rexx syntax in any of them?

In what way do You need support ?  There are support for syntax
highlightning for ASM-S/370.
