Nicolas wrote a nice set of instructions for translating at

I maintain the Spanish translation of interface using Tortoise SVN for getting 
the files from 
SVN server at SourceForge, poEdit for the translation (both under Windows) and 
then 
Tortoise SVN  again for uploading --comitting-- the updated files.
It's an easy process, although a great lot of things to translate now!

As for starting a new language (pt-BR), I think you can do it as explained at 
the URL above, 
or Nico will know how to do that.
