Yes, you should be able to just copy everything over, then use the Startup Disk control panel to tell your Mac to start up off the other disk. Reboot and you should be running off the new disk. You can tell which disk the computer is running off by looking at which disk icon is in the very top right - if it is you're new disk, then you're all set.
