There are think kind of commands and they are the same as sqlite, so
if you need to diagnose/repair a database just send this SQL command:
PRAGMA integrity_check; (http://www.sqlite.org/pragma.html)
to defragment just send:
VACUUM (http://www.sqlite.org/lang_vacuum.html)
Does the database never get corrupted with the time and the
successive INSERT, UPDATE and DELETE calls?
It is very unlikely because the database is fully ACID compliant.

Does the BACKUP feature create an entirely rebuilt file ? Or is it
a binary copy of the original file — with the same corruptions if
there are any ?
It is a binary copy ... this is the fastest way, but the database is
sanity checked before the backup.
Are there some limitation such as number of records within a table?
No, you are only limited by the database size: 2 terabytes.
I can't find any White Papers on RS site, nor users' feedback in
the nug list...
In the meantime you can just ask me :-)
