Will adding a SATA card be enough or will the bios potentially be a
problem also?
In a word - No.  The sata has its own drive controler built in and does
not rely on bios to function.

I'll want to keep the "server" PC running 24/7 so I don't have to boot
up every time I want to play music...will that be a problem or lead to
early HD death?
Again - no.  Look into the S3 power function in your bios.
(http://tvtool.info/FlashHelp/S3_Power_mode.htm)  It shuts down all but
the memory and your pc will "wake" at the press of a keyboard key or
when you turn on your Squeezebox if your box is wired.  If its wireless
you will need to wake the pc first.
