The biggest killer of HDD is excessive heat. What is worse, is excessive heat followed by cooling, and cycling that pattern over and over. I actually believe find that given you have adequate airflow in the case, leaving the PC powered up is better for the overall life of the HDD. I use an interal SATA card capable of doing RAID5 and run 4 disks in that configuration.
