No, no, no.

There is no difference between ripping straight to MP3 and ripping to
FLAC and then to MP3.

If you use the same MP3 encoder for both processes, with the same
settings then the resulting MP3 files will be absolutely identical. How
good the listeners ears are will not change the fact that the files will
be identical.
