One have all the domain accounts and the other is the default smtp
server for all domain users. The problem is that some messages from
domain users to domain users are getting 4.4.2 error (connection died).
Bothe servers are on the same LAN.

No, I did. the qmail-queue patch only mods 2 files and the config for it
is also by itself.
qmail-queue will slow down mail processing (did in my case) so if its a
medium/high volume smtp server then you better plan for some additional
bogomips to fire off the scanning.
