Mhhh... MasterIncludesPlugin is not from me. I merely created that
TiddlySpace as I found the previous ways of getting to understand what
it's about and how it works somewhat tedious. I have now put a notice
at the beginning of this space, which makes this clear.

As for externalizing plugins or contents with it ...I really haven't
tried that yet as I primarily wanted to have an ability to edit my
plugins in my editor while having them automatically loaded into my
DevWiki.

re a+c) I haven't tried it myself, but I would assume that
transclusion works, otherwise it would seem quite a shortcoming.

re b+d) Editing does not. Afaik, you have to edit remote tiddlers in
their original document.

As for replacing...

<<tiddler [[TiddlerName##TiddlerName]]>>

with...

<<tiddler [[TiddlerName##Content]]>>

...that seems in no way any more complicated than replacing...

!TiddlerName

with...

!Content

...although it would probably apply to a whole lot more than just your
glossary entries ...namely all the places where you are referring to
them, possibly some inline sliders, etc...
