They send POT files for stuff that isn't translated at all and PO files
for stuff that has some th translations. You probably want to see what
is the most important for you to do. The ISO codes, especially for
countries and languages in your region are certainly worth doing, we use
them in Pootle.

Changes you make outside of Pootle do require a restart.

I'd suggest doing this.  You can safely make your pot files into th.po
and simply copy any .th.po they send you into your th directory.  Doing
it this way you don't need to put anything into templates, you also
won't need to update from within Pootle.

If your PO files follow that format then no.  You can probably create a
nice script to mail these if you use xdg-email and attach all the files
and send to [email protected]
