Like Chris said :) I for one use Mochikit in all my Ajax related tasks, it makes javascript suck so much less! I know for sure most people doing serious Ajax/EventHandling out there use Mochikit :-) (a troll is hiding inside this sentence will the reader avoid it?) +1 on Mochikit for js programing. This does not mean that I don't use other libs for display effects... ever looked at qooxdoo? (http://qooxdoo.org/) Genshi + Tosca* + SQLAlchemy will all stay in my toolbox. So +1 for all of them :) I also like cherrypy and its object publishing a lot.
