I don't believe they've ever made an attempt to stop the Standing O. That's a holdover from the Letterman days, where they also did it when Dave came out on stage. It was the Colbert Report/Jerry Stringer "Steven" chant that they put the kibosh on (although that still occasionally happens with some disobedient audiences).
And yes, that was the first Leno-esque audience high-fiving that I've seen.
