As a general rule, you should always unplug any computer before inserting a new 
card. It's more critical on some machines than on others, but leaving it 
plugged in is inviting possibly shorting out both the card and the motherboard 
if your hand slips and something touches where it shouldn't.  Not to mention 
the possibility of your getting a shock.

As far as the trickle voltage you refer to, to the best of my knowledge, that 
serves to allow the computer to respond to the keyboard power switch, and 
doesn't have anything to do with the pram.
